Glycine: The Underrated Sleep Amino Acid

Three grams before bed, measurable effects on sleep onset and next-day alertness, and a second life as the limiting amino acid for collagen and glutathione.

The Short Answer

Glycine is the smallest amino acid, an inhibitory neurotransmitter in the brainstem and spinal cord, a co-agonist at NMDA receptors, and the most abundant amino acid in collagen, where it occupies every third position. Trials at 3 grams before bed report shortened sleep onset latency on polysomnography, improved subjective sleep quality, and improved next-day alertness and reduced fatigue, with the proposed mechanism being peripheral vasodilation that accelerates the core temperature fall sleep onset requires. Separately, dietary glycine intake is frequently below the amount needed for optimal collagen and glutathione synthesis, which is a distinct and under-recognised case for supplementation.

The Sleep Evidence

The relevant trials come mostly from a Japanese research group and are small but methodologically interesting because several used objective measures.

In participants with sleep complaints, 3 grams of glycine before bed shortened polysomnographically measured sleep onset latency and time to slow wave sleep, without altering sleep architecture proportions. Subjective sleep quality improved.

In a partial sleep restriction protocol, glycine reduced next-day fatigue and daytime sleepiness and improved performance on a psychomotor vigilance task relative to placebo. That combination, better sleep onset without next-day sedation, is unusual and is the practical selling point.

Mechanistic work in rats found that glycine administration lowered core body temperature through peripheral vasodilation, mediated by NMDA receptors in the suprachiasmatic nucleus. Since sleep onset depends on core temperature falling, accelerating that fall is a coherent route to faster onset.

The literature is small and concentrated in one group, which is a limitation. The doses are cheap, the compound is a normal dietary constituent, and the effect is fast enough to test personally within days.

The Collagen and Glutathione Case

This is the less-discussed and arguably more interesting half.

Glycine is conditionally essential: the body synthesises it, and synthesis capacity appears insufficient to meet full demand. Analyses of glycine metabolism estimate a shortfall of several grams per day between what is synthesised plus typically consumed and what collagen synthesis alone would require.

Collagen is roughly one third glycine by residue count, which makes it the single largest demand. Glutathione, the main intracellular antioxidant, is a tripeptide of glutamate, cysteine and glycine, and glycine availability has been shown to limit glutathione synthesis in older adults, where combined glycine and cysteine supplementation restored glutathione concentrations and improved several markers of oxidative stress and mitochondrial function.

Modern dietary patterns are relevant here. Muscle meat is low in glycine relative to connective tissue, skin and bone, and diets that exclude the collagen-rich parts of animals supply less glycine than historically typical patterns did. This is one of the few nutritional arguments for eating nose-to-tail that has a specific biochemical basis rather than a rhetorical one.

Dose, Timing and Sources

  • 3 g before bed. The sleep trial dose. Glycine is sweet-tasting and dissolves readily in water.
  • 3 to 10 g per day. The range used for metabolic and glutathione purposes, often divided.
  • 15 g per day and above. Used in some clinical research contexts, including schizophrenia adjunct trials at much higher doses, without significant toxicity.

Food sources. Gelatin and collagen are roughly a quarter to a third glycine, so bone broth, skin, connective tissue and collagen supplements all supply it in gram quantities. A collagen supplement taken for skin or joints is, incidentally, a glycine supplement.

Combination with cysteine. For glutathione purposes, glycine alone is insufficient: the trials that restored glutathione in older adults combined glycine with N-acetylcysteine. Cysteine availability is the other limiting factor.

Safety. Glycine has an excellent safety record with no established upper limit for dietary use and a long history of use as a food additive. Reported effects at high doses are mild gastrointestinal upset. As an NMDA co-agonist it interacts theoretically with medications acting on that receptor, including clozapine, where glycine has been reported to reduce efficacy. Anyone with significant kidney impairment should regard gram-level amino acid dosing as a clinical question.

Where Glycine Fits Among Sleep Compounds

Compared with the alternatives, glycine has specific advantages: it works within days, it is inexpensive, it has an objective polysomnographic result behind it, it does not produce next-day grogginess, and it is a normal dietary constituent rather than a pharmacological agent.

Its limitations are equally clear: the evidence base is small and concentrated, the effect is on sleep onset rather than maintenance, and it is not a sedative, so someone expecting to be knocked out will be disappointed.

In practice it pairs sensibly with magnesium glycinate, which supplies some glycine already, and with the behavioural layer that outperforms both. For the person whose complaint is specifically that they take too long to fall asleep and who does not want next-day sedation, it is among the more sensible first things to try.

The Longevity Angle

A final thread worth noting, because it connects glycine to the wider content of this Journal.

Glycine supplementation extended lifespan in rats in work from a multi-site programme, and glycine restriction has effects in the opposite direction in some models. Proposed mechanisms include methionine restriction mimicry, since glycine clearance of excess methionine through the glycine N-methyltransferase pathway lowers methionine availability, and improved glutathione synthesis.

The methionine connection is the more interesting one. Methionine restriction extends lifespan robustly in rodents, and glycine may achieve part of the same effect without dietary restriction, since it accelerates methionine disposal. This is animal work, and it is a mechanistically coherent reason for interest beyond sleep.

Nothing here justifies claims about human longevity. It does mean that a cheap, safe, dietary amino acid with a sleep effect also sits on a pathway the field takes seriously, which is more than most sleep supplements can say.
